Tissue factor pathway inhibitor

protein that is predominantly absorbed at the endothelia and plays an important role in clotting.TFPI binds factor Xa to a complex that antagonizes the tissue thromboplastin factor VIIa complex. In addition, factor X activation is stopped. The molecular weight is between 34-40 kD. The half-life is 100 minutes.
In plasma, TFPI is bound to lipoproteins. Only about 10% is present in free form. The synthesis of the tissue factor pathway inhibitor takes place in the endothelium.

A recombinant TFPI (tifacogin) was tested as an anticoagulant for the therapy of sepsis. It was not used in the clinic because there was a significant increase in the bleeding rate.
