The highest expressions of CD34 are found on primitive stem cells. They gradually disappear during differentiation and maturation of the precursor cells. About 60% of all acute B-lymphoid leukemias and 40% of acute myeloid leukemias (AML) and up to 5% of acute T-lymphoid leukemias express CD34.

CD 34 is the best marker of dermatofibrosarcoma protuberans. CD34 also marks numerous soft tissue tumors: solitary fibrous tumor, Kaposi's sarcoma, angiosarcoma, neurogenic tumors, spindle cell lipoma, epithelioidal sarcoma.
